What Is Datroway Used For?

Datroway (datopotamab deruxtecan-dlnk) is an antibody-drug conjugate (ADC) that combines an antibody targeting TROP2 (a protein found on the surface of many solid tumors) with a cytotoxic topoisomerase I inhibitor payload, linked by a cleavable linker. After binding to TROP2-expressing cancer cells, it is internalized and releases the cytotoxic payload, causing DNA damage and cell death. It is indicated for the treatment of:

  • Adult patients with unresectable or metastatic, hormone receptor (HR)-positive, HER2-negative breast cancer who have received prior endocrine-based therapy and chemotherapy for unresectable or metastatic disease.
  • Adult patients with locally advanced or metastatic EGFR-mutated non-small cell lung cancer (NSCLC) who have received prior EGFR-directed therapy and platinum-based chemotherapy. This indication is approved under accelerated approval based on response rate and duration of response.
  • Adult patients with unresectable or metastatic triple-negative breast cancer (TNBC) who are not candidates for PD-1/PD-L1 inhibitor therapy.

Datroway is a prescription anticancer medicine and must be administered under the supervision of a qualified medical oncologist.

Recommended Dosage

Datroway is given as an intravenous infusion at a recommended dose of 6 mg/kg of body weight, once every 3 weeks (21-day cycle), up to a maximum of 540 mg for patients weighing 90 kg or more. Treatment continues until disease progression or unacceptable toxicity. Premedication is recommended before infusions to help prevent infusion-related reactions and nausea/vomiting.

Doses may be reduced, delayed, or permanently discontinued based on the severity of interstitial lung disease/pneumonitis, ocular adverse reactions, stomatitis, or other toxicity. The exact dose, schedule, and duration of therapy must be individualized by the treating physician according to the full prescribing information.

Datroway Warnings and Precautions

  • Treatment with Datroway can cause serious, potentially life-threatening or fatal lung inflammation. Report new or worsening cough, shortness of breath, fever, or respiratory symptoms immediately. Treatment may be withheld if suspected and permanently discontinued for confirmed Grade 2 or higher cases.
  • This medicinal product may cause eye problems, including dry eye, keratitis, blepharitis, conjunctivitis, increased tearing, and blurred vision. Eye examinations are recommended before treatment and periodically thereafter. Use preservative-free lubricating eye drops and avoid contact lenses during treatment.
  • Datopotamab deruxtecan-dlnk can commonly cause mouth ulcers and sores, which may sometimes become serious. Maintaining good oral hygiene is recommended. Patients should promptly inform their healthcare provider about mouth pain, sores, or difficulty eating during treatment.
  • This therapy may reduce white blood cells, red blood cells, and platelets and may affect liver enzymes or other laboratory values. Regular blood tests are recommended during treatment to monitor blood counts, liver function, and other relevant laboratory parameters.
  • Infusion-related reactions may occur during or shortly after Datopotamab deruxtecan-dlnk administration and can include bronchospasm. Healthcare professionals will monitor patients during and after each infusion to identify reactions promptly and provide appropriate medical management when necessary.
  • This medicine may harm an unborn baby. Females of reproductive potential should use effective contraception during treatment and for 7 months after the last dose. Males with partners of reproductive potential should use contraception during treatment and for 4 months afterward.
  • Datroway is classified as a hazardous drug and requires special handling. It should be prepared, handled, and disposed of by trained healthcare professionals according to applicable safety procedures to minimize occupational exposure and protect healthcare personnel.
